Abstract

Aims The aim was to perform a meta-analysis on the miRNA expression profiling studies in diabetic nephropathy (DN) to identify candidate diagnostic biomarkers. Methods A comprehensive literature search was done in several databases and 53 DN miRNA expression studies were selected. To identify significant DN-miR meta-signatures, two meta-analysis methods were employed: vote-counting strategy and the robust rank aggregation method. The targets of DN-miRs were obtained and a gene set enrichment analysis was carried out to identify the pathways most strongly affected by dysregulation of these miRNAs.

Abstract

Acute kidney injury (AKI) is one of the most common health-threatening diseases in the world. There is still no effective medical treatment for AKI. Recently, Mesenchymal stem cell (MSC)-based therapy has been proposed for treatment of AKI. However, the microenvironment of damaged kidney tissue is not favorable for survival of MSCs which would be used for therapeutic intervention. In this study, we genetically manipulated MSCs to up-regulate lipocalin-2 (Lcn2) and investigated whether the engineered MSCs (MSC-Lcn2) could improve cisplatin-induced AKI in a rat model. Many studies have investigated misregulation of miRNAs relevant to multiple sclerosis (MS) pathogenesis. Abnormal miRNAs can be used both as candidate biomarker for MS diagnosis and understanding the disease miRNA-mRNA regulatory network. In this comprehensive study, misregulated miRNAs related to MS were collected from existing literature, databases and via in silico prediction. A multi-staged data integration strategy (including the construction of miRNA-mRNA regulatory network and systematic data analysis) was conducted in order to investigate MS related miRNAs and their regulatory networks. Results: the pPICZ-TF construct contain a full length cDNA of the rabbit tissue factor gene which cloned downstream the AOX1 promoter and upstream the His-tag. The pPICZ-TF construct inserted in downstream the AOX1 gene on chromosome 4 and Phenotype of recombinant strains was Mut+. Expression of rRTF (43KD) was confirmed by performing of SDS-PAGE and Western Blot. Empty liposomes were made in Rotary evaporator and their sizes were decreased by extrusion method. For obtaining optimal activity purified rRTF was incubated with empty liposomes. Then the activity of the relipidated rRTF was confirmed by PT test. Abstract

Methods: ADSCs were isolated from three liposuction samples and their conditioned medium was collected. Skin biopsies were isolated from six patients and their fibroblasts were isolated. These cells were cultured in 100% of ADSC-CM. Expression of three HA synthase and two hyaluronidase genes were assessed with the use of real time PCR, with ELISA and final amounts of secreted HA were measured in the treated and control groups.Results: Gene expression of two HA synthase genes HAS1 and HAS2 were upregulated in treated samples compared to the control group.
